AssessmentsPlus™

AssessmentsPlus™ empowers GPX users to efficiently build high-quality PBPK/PBBM models using embedded expert knowledge from highly experienced PBPK/PBBM mechanistic modeling scientists. AssessmentsPlus™ provides the Compound Assessment, to advise the user before modeling, and the Simulation Assessment to provide an in-depth understanding on the absorption, distribution, metabolism and excretion driving the pharmacokinetic profile. If observed plasma concentration vs time data is available, the Simulation Assessment also provides recommendations for improving the fit of the model.

In the Compound Assessment, the compound is classified based on a series of defined attributes (e.g., solubility, permeability, lipophilicity, extent of clearance, etc.). The compound properties are paired with physiological, dosing, and pharmacokinetic information to enhance the specificity of advice. These inputs are then fed into a series of proprietary decision trees covering absorption, distribution, metabolism and excretion to procedurally generate the feedback that will be returned to the user.

The Simulation Assessment runs the selected simulation in real-time, using the simulation inputs and outputs to conduct an analysis of the simulation results, and provides qualitative and quantitative feedback on absorption, distribution, metabolism and excretion. For comparison to observed exposure data, the assessment highlights any non-alignment and provides recommendations regarding which parts of the model should be examined and how they should be adjusted.

As part of the GastroPlusGPT™ suite of AI-powered web apps, the AssessmentsPlus™AI module provides an optional enhancement for both the Compound Assessment and Simulation Assessment. This app leverages our tuned LLM, trained on GastroPlus® documentation, to organize the feedback and provide additional recommendations supported by the Assessment data. Additional plots and tables are used to enhance understanding and explain the pearls of wisdom from GastroPlus®. The recommendations are clearly grouped together and include the ability to interact with the chatbot to answer any questions about the Assessment and generate clear step-by-step instructions for implementation in GastroPlus®.
